There is no single answer, because the detection window depends on the body fluid being tested and on which substance the test looks for. Nicotine itself clears within a day or two. Cotinine — the primary metabolite produced when your liver breaks down nicotine — lasts several times longer. Why labs measure cotinine, not nicotine

Nicotine is metabolized so quickly that it is nearly useless for detecting past use. Within two hours, half of a dose has already been converted by the liver enzyme CYP2A6 into cotinine. Because cotinine has a half-life of about 16 hours — roughly eight times longer than nicotine — it provides a much wider and more reliable detection window. Cotinine is also more stable in stored samples, which is why the major reference laboratories (Quest, LabCorp, and ARUP) build their nicotine screening panels around cotinine rather than nicotine.

There is one caveat: cotinine does not distinguish between nicotine from cigarettes and nicotine from nicotine replacement therapy. A person using nicotine gum or a patch will produce the same cotinine result as a smoker. That matters for employment screening — if you use NRT, you need to tell the tester before the sample is collected.

Detection windows are averages. Your personal window depends on these variables.

1. How much and how often you use nicotine. The single biggest factor. Heavy, chronic users accumulate cotinine in their tissues; after quitting, those stores drain slowly, which is why heavy smokers can test positive for two to three weeks while an occasional user clears in days.

2. Your metabolic rate and liver enzymes. People with faster CYP2A6 activity convert nicotine to cotinine and clear both faster. This genetic variation is why two people with identical habits can have very different detection windows.

3. Age and body composition. Older adults tend to clear nicotine and cotinine more slowly. Cotinine is stored in fat tissue, so body composition influences how long it lingers after the last dose.

4. Hydration and kidney function. Cotinine is excreted in urine, so well-hydrated kidneys clear it somewhat faster. The effect is modest — hydration cannot flush you clean, but chronic dehydration can extend the window.
